Abstract

The increasing prevalence and dissemination of multidrug-resistant bacteria represent a serious concern for public health. Aeromonas caviae is a pathogenic microorganism that causes a wide spectrum of diseases in fish and humans and is often associated with aquatic environments and isolated from foods and animals.

Here, we present the isolation and characterization of the V15T strain isolated from a drinking water storage tank in Rio de Janeiro, Brazil.

The V15T strain has a genome length of 4,443,347 bp with an average G + C content of 61.78% and a total of 4028 open reading frames. Its genome harbors eight types of antibiotic resistance genes (ARGs) involving resistance to beta-lactamases, macrolides, and quinolones. The presence of blaMOX-6, blaOXA-427/blaOXA-504, and mutations in parC were detected.

In addition, other ARGs (macA, macB, opmH, and qnrA) and multidrug efflux pumps (such as MdtL), along with several resistance determinants and 106 genes encoding virulence factors, including adherence (polar and lateral flagella), secretion (T2SS, T6SS), toxin (hlyA), and stress adaptation (katG) systems, were observed.

The genome sequence reported here provides insights into antibiotic resistance, biofilm formation, evolution, and virulence in Aeromonas strains, highlighting the need for more public health attention and the further monitoring of drinking water systems.

Also, the results of physiological and phylogenetic data, average nucleotide identity (ANI) calculation, and digital DNA–DNA hybridization (dDDH) analysis support the inclusion of the strain V15T in the genus Aeromonas as a new subspecies with the proposed name Aeromonas caviae subsp. aquatica subsp. nov. (V15T = P53320T).

This study highlights the genomic plasticity and pathogenic potential of Aeromonas within household drinking water systems, calling for the revision of water treatment protocols to address biofilm-mediated resistance and the implementation of routine genomic surveillance to mitigate public health risks.

When does it make sense to use an antiviral to treat H5N1 flu in a dog or cat?

Use of an antiviral makes sense for early treatment of known or high risk cases of H5N1 influenza where there’s a concern for development of serious disease (i.e. any infected cat, and probably infected dogs) AND when the animal can be properly treated AND when the animal can be kept isolated during and shortly after the treatment period.

Basically my two main questions are: do they need it? and am I confident the animal won’t be able to infect another individual (human or animal)? If I can comfortably say yes to both of those, I think it’s reasonable to use an antiviral.

Example 1: An infected cat in a household or veterinary clinic

Yes. We can properly treat, isolate, monitor and test the cat appropriately.

Example 2: An infected cat that goes outside

No, unless the cat can be kept inside during the treatment and monitoring period. I don’t want to risk an antiviral-resistant flu strain developing and then the cat spreading it to other cats, or worse, birds.

Example 3: A potentially infected backyard chicken

No (or hell no). These are livestock, so they are approached differently (and in Canada poultry infected with H5 flu must be culled). Poultry are highly susceptible to H5N1 influenza, and can clearly infect people. Also, an antiviral is probably too little, too late for a species that is so susceptible.

DJ, We need to reduce potential hosts. NL has over 100 million farm animals (on a population of 18 million people). Less meat consumption-better price for farmers-better quality of life for (farm)animals !

Anecdotal report: I've heard from a reliable friend who is fluent in pandemics that a contact at CDC has said that the recent 11 (now 12) herds across 3 states (CA, NV, ID) have resulted in workers who are symptomatic. This is not surprising. It's my belief that a great deal of Tamiflu has been handed out since the great rush of workers with symptoms occurred and then very suddenly subsided. The news here is that the CDC contact stated that RFKjr has directed that the symptomatic workers should not be tested. Data is clearly the first casualty of the coming pandemic. We'll be willfully flying blind.

Aeromonas caviae is a pathogenic microorganism that causes a wide spectrum of diseases in fish and humans and is often associated with aquatic environments and isolated from foods and animals. Here, we present the isolation and characterization of the V15T strain isolated from a drinking water storage tank in Rio de Janeiro, Brazil. The V15T strain has a genome length of 4,443,347 bp with an average G + C content of 61.78% and a total of 4028 open reading frames. Its genome harbors eight types of antibiotic resistance genes (ARGs) involving resistance to beta-lactamases, macrolides, and quinolones. The presence of blaMOX-6, blaOXA-427/blaOXA-504, and mutations in parC were detected. In addition, other ARGs (macA, macB, opmH, and qnrA) and multidrug efflux pumps (such as MdtL), along with several resistance determinants and 106 genes encoding virulence factors, including adherence (polar and lateral flagella), secretion (T2SS, T6SS), toxin (hlyA), and stress adaptation (katG) systems, were observed. The genome sequence reported here provides insights into antibiotic resistance, biofilm formation, evolution, and virulence in Aeromonas strains, highlighting the need for more public health attention and the further monitoring of drinking water systems. Also, the results of physiological and phylogenetic data, average nucleotide identity (ANI) calculation, and digital DNA–DNA hybridization (dDDH) analysis support the inclusion of the strain V15T in the genus Aeromonas as a new subspecies with the proposed name Aeromonas caviae subsp. aquatica subsp. nov. (V15T = P53320T). This study highlights the genomic plasticity and pathogenic potential of Aeromonas within household drinking water systems, calling for the revision of water treatment protocols to address biofilm-mediated resistance and the implementation of routine genomic surveillance to mitigate public health risks. Over the previous 6 months, China has reported an additional 13 cases (see here, here, and here), with well over 100 reported over the past decade (see FluTrackers list). Most cases are mild, and seroprevalence studies suggest many cases may go undetected. H9N2 is poorly controlled in Chinese poultry, despite the use of vaccines (see J. Virus Erad.: Ineffective Control Of LPAI H9N2 By Inactivated Poultry Vaccines - China), which has led to the creation and spread of numerous of genotypes. H9N2 also reassorts with, and often enhances, other novel influenza viruses (including H7N9, H5N1, and H5N6), making it an important co-conspirator (see Vet. Sci.: The Multifaceted Zoonotic Risk of H9N2 Avian Influenza). Seven years ago, in EID Journal: Two H9N2 Studies Of Note, we looked at two reports which suggested that H9N2 continues to evolve away from current (pre-pandemic and poultry) vaccines and is potentially on a path towards better adaptation to human hosts. While LPAI H9N2 is admittedly not at the very top of our list of pandemic concerns, the CDC has 2 different lineages (A(H9N2) G1 and A(H9N2) Y280) on their short list of influenza viruses with zoonotic potential (see CDC IRAT SCORE), and several candidate vaccines have been developed. I don’t want to risk an antiviral-resistant flu strain developing and then the cat spreading it to other cats, or worse, birds. Example 3: A potentially infected backyard chicken No (or hell no). These are livestock, so they are approached differently (and in Canada poultry infected with H5 flu must be culled). Poultry are highly susceptible to H5N1 influenza, and can clearly infect people. Also, an antiviral is probably too little, too late for a species that is so susceptible. DJ, We need to reduce potential hosts.
